No, you use water to calibrate it according to the instructions.
Water and sugar, you need to set different gravities, using a hydrometer or refractometer.

Yes but then you need to read a hydrometer to calibrate it! Well, I did my first calibration with a refractometer which seemed to go quite well but I couldn't get consistent readings the second time.

Just a word of caution that on the iSpindel firmware list the "Latest release" is 5.9.0 but this doesn't work with Ubidots due to a bug that I've wasted all evening tracking down.
