At what point should you take opening gravity?

I have a Young's Bitter which dropped to 1.006 as per instructions but forgot to take opening gravity so I don't know what it's alcohol content is.

On my next brew, i'd like to know. Do you take it prior to adding yeast? Does it make a difference?

This is the way i estimate alcohol content and it gives me an idea of what FG to expect. Sorry its in metric but google can easily convert to imperial.... :D
